The single was released in August of 1963 with "Blowin' in the Wind" on the A-side and "Don't Think Twice, It's All Right" on the B-side. Both songs were contained on Dylan's second studio album, "The Freewheelin' Bob Dylan." The other single released from this album featured "Mixed-Up Confusion" on the A-side and "Corrina, Corrina" on the B-side.

Suze Rotolo is pictured walking with Dylan on the cover of "The Freewheelin' Bob Dylan" album. She first met Dylan in July of 1961 at a Riverside Church folk concert. In June of 1962, she went to study at the University of Perugia in Italy. About the time this song was written, she decided to prolong her stay. According to Songfacts, Suze Rotolo's obituary in The Guardian, and several other sources, this separation is thought to have inspired several of Dylan's songs in addition to "Don't Think Twice, It's All Right," including "Boots of Spanish Leather," "One Too Many Mornings," and "Tomorrow is a Long Time."
