Giles is the neighbor to whom Elisa confides in and listens to for advice. During the movie Giles discovers the creature at the apartment eating one his cats, startles it, causing it to injure him. As the movie builds the connection between the creature and Elisa grows stronger. They are able to relate through non-verbal communication and become enamored with each other. Hoffstetler is a Soviet spy who is employed as a scientist at the laboratory during the Cold War era of the 1960s. He is intent on keeping Amphibian Man alive for further study and is opposed to any experimentation that involves vivisection. Despite also receiving orders from the Soviets to kill the creature, he resists and agrees to help Elisa free it.

During his effort to find the escaped creature, Strickland follows Mosenkov, also known as Hoffstetler, who is required to meet up with his superiors to explain why the creature has gone missing. At the rendezvous, Mosenkov is shot by one of his handlers and in turn Strickland shoots the entire group.

While Mosenkov lays dying, Strickland tortures him until he reveals that Elisa and Zelda had freed the creature.

After having received information from Mosenkov, Strickland rushes to the apartment building to confront Elisa and Zelda. He finds Zelda and threatens her, causing her husband Brewster to 'spill the beans'. Strickland then enters Elisa's unit and finds a note indicating that the creature is soon to be released into the canal.

During the closing scenes at the canal, Elisa and Giles prepare to usher off the creature back to its watery home when Strickland appears and shoots both Elisa and the creature. Both appear to die but the creature heals himself, grabs Elisa, and the two jump in the canal where he heals her as well and provides her with gills. Presumably, they live together happily ever after underwater.
