"Machete" is the story of a Mexican federale who vows revenge against the American hit men who wronged him. Alongside Father Benicio Del Toro (Cheech Marin), he destroys all those who get in his way, while getting a chance with the ladies. "Machete" plays during the actual "Grindhouse" film, but after the opening credits. Since it was made by "Robert Rodriguez", it is somewhat packaged with the film. Danny Trejo plays Machete. The other three mock trailers are played between "Planet Terror" and "Death Proof" and are made by Eli Roth, Rob Zombie, and Edgar Wright. Each of them explores different genres of horror, but each one is no less than what you'd see in an actual grindhouse.

The movie opens with Jungle Julia and her friends, Arlene and Shanna, as they get together for a day and night in Austin's bars. When they drive to their first bar, Guerro's, they pass by several billboards erected to advertise Julia's radio show. They also discuss Arlene's new boyfriend, their upcoming weekend at the lake together, and the fact that they want to score some marijuana. When the three of them stop along the way at a Mexican restaurant for lunch, Arlene notices a souped up, black Chevy Nova with a skull and crossbones on the hood slowly drive by. Little do she and her friends know that the man inside has been following them for quite some time.

One of the five faux trailers featured alongside "Planet Terror" and "Death Proof" won a contest held by Robert Rodriguez, that would put some amateur filmmakers in the spotlight. The winners were a couple of guys from Nova Scotia, who had their film preview for "Hobo with a Shotgun" showcased. Their proposed idea: a hobo takes the law into his own hands, killing criminals and evil-doers with a shotgun along the way.

In the opening scenes of the movie, Cherry is doing her go-go dance routine on stage, but she ends up crying by the end. When she's in the dressing room, her boss tells her that she can't keep crying during the dance because the go-go is supposed to be a happy dance (as he puts it, it's 'go-go', not 'cry-cry'). She decides to get rid of that useless talent to find a use for her others, so she flips him the bird, takes her money, and leaves to club. While she walks down the road, three trucks nearly hit her on their way to the military base. One of these contains scientist, Abby, as he goes to meet some people for his 'specimens'.

On the night of the horrible toxic chemical release, Cherry Darling quit her go-go dancing job in town, in order to put her unusual talents to good use (possibly a stand-up comedy career that Wray finds misfitting). Ironically, after she is attacked by the living dead, she finds herself missing a leg. Wray takes the opportunity to attach a table leg to her thigh, and later, a machine gun.

While at a diner, Abernathy tells the other two girls they're with an anecdote about filming a movie ("Three Kicks to the Head Part III") in the Philippines. Abernathy goes on to illustrate how she almost fell into a seven foot concrete ditch full of rats and broken bottles, and how bad she felt about herself when Zoe fell in and came out with no broken bones at all.

During their meeting, Lt. Muldoon realizes that Abby has been holding extra canisters of the dangerous chemical DC2, so he decides to take it all. Abby tries to retaliate, so he shoots the canisters, releasing the gas into the air. This inadvertently saves the army men, but all of the people killed in the battle around them have turned into zombie-like creatures. Abby makes it out alive, but a sicko is attacking him from on the roof of the car. Abby passes by a gas station and The Bone Shack on his way down the road. It is here where Cherry reunites with her former lover, El Wray. She discusses her future with him and the fact that she no longer wants to be a go-go dancer, but a comedienne instead (a fact made even more ironic when she loses her leg).

At a nearby military base, a businessman named Abby nearly hands sealed containers of DC4 over to Lt. Muldoon, but when a traitorous plot is revealed, he shoots one of the units and releases the gas into the air. It is later revealed that Abby is a scientist who knows the secrets behind the deadly chemical and that all of the military men (including Muldoon) are infected by a controlled source of the bacteria. Muldoon also reveals that he murdered Osama Bin Laden. Both are killed shortly after.

Tammy stops at The Bone Shack before Cherry does, in order to fix her car which is overheating (due to a stripped radiator cap). She tells JT that she needs to get to the town urgently. He asks if it's to save lives, and she says yes. Meanwhile, Doctors William and Dakota Block are waking up for their night shift at the local hospital. Dakota mysteriously talks with Tammy over a palm pilot while her son eats his 'breakfast' to say that she'll be taking her son with her. Evidently, she's planning an escape. They make their way to the hospital where William's first patient is a local fisher with an odd bite on his arm. He and another doctor investigate the wound, find that the man has a fever of 105, and that he has tons of abscesses on his tongue. He tries to pop one, but it bursts.

Stuntman Mike ends up buying a beer for both Arlene and Jungle Julia and surprising them with the poem. Julia quickly stands up for her friend (realizing it was a stupid thing to do) and tells him that she has already given the lap dance to someone at another bar, but Mike says that she's lying. He asks Arlene why she's so afraid of him, and she says that it's his car. He quickly charms her over though, and exposes her bruised ego since she never had any guys recite the poem for her, and she heads into the bar to give Mike his lap dance. Unfortunately, the theatrical version of the film is missing the entire scene, and the audience is quickly taken to the scene where everyone is leaving the bar.

Another major character in the story is Dr. Dakota Block, whose husband is also a doctor in the nearby hospital. The two are unhappy with their marriage, and Dakota is ready to sneak away with her lesbian lover, Tammy, who was killed by zombies on the way into town. When her husband finds out, he numbs her and hides her away, but she jumps through a hospital window, steals a car (breaking her numb hand in the process) and takes her son (while fleeing from two angry babysitters who smash her car). When she heads to her dad's house, her young son accidentally shoots himself in the car and her husband has become another of the undead creatures. Her father later comes to her rescue when she's cornered by her husband, inside the cockpit of an abandoned plane at the military base.

At the Texas Chili Parlor, Warren joins the small crowd around the girls and, referring to Jungle Julia's statement ("Them's the rules baby, Warren says it - we do it"), orders them to take green shots he later reveals to be Chartreuse. For anyone who doesn't know, Chartreuse is a green or yellow French liqueur (about 100 or 110 proof). It is one of the handful of liquors which continues to age and improve in the bottle, and it also gives it's name to the blinding neon green/yellow.

When Dr. Block sends for his wife to help Joe, she brings what she calls her three friends. When she arrives, Joe's arm is nearly completely covered by the sores and William fears they may need to cut his arm off immediately to prevent further spreading of the infection. This is where Dakota comes in. Her first 'friend', the yellow needle, is used to take the sting off of it. The second, blue needle, doesn't have any feel to it, and after the third, she says, they'll never see her again. Meanwhile, Tammy's car dies on the road to town, two miles outside the military base. As she tries to flag down a car for help, she is promptly attacked by sickos and killed. El Wray and Cherry watch to see her dragged off the road. They naturally assume it's roadkill.

When everyone leaves the bar, Stuntman Mike shows Pam to his car. Although she is nervous that it may be dangerous, he assures her that the entire machine is 'death proof', since it was made to withstand stunts and crashes. She finally accepts the reassurance and hops into the empty passenger seat, which is little more than a metal seat surrounded by reinforced steel bars. As they head out of the parking lot, Stuntman Mike asks which way she needs to go to get home. When she says 'right', he is disappointed, as she'll soon catch on to the fact that they won't be going in the right direction. He quickly speeds up, spins the car, and slams the brakes, killing her when she impacts with the windshield. Of course, he isn't harmed- the driver's seat is the only 'death proof' part of the car. He speeds in the direction that Jungle Julia and her group went and passes them on the way. Unfortunately, everyone in Jungle Julia's car (driven by her dealer, Lanna Frank) is hammered. They don't even notice when Stuntman Mike spins around up ahead, turns off his headlights, and crashes into their car head-on. All four girls inside are brutally murdered.

Hiding away at JT Hague's steakhouse, the survivors fight off the zombies and relax. Cherry realizes that Wray was going to propose to her and they have a passionate time together. A missing reel breaks up the story, but shortly afterwards, the restaurant is on fire and all of the survivors have gathered inside. They decide to make their way to the Mexican border and they make their way there, but they're stopped by the military who place them in prison. Cherry and Dakota are taken away, but are saved at the last minute by Wray and Abby. They all manage to find a helicopter, and with tons of guns, fight off the military undead. Wray is shot and killed, but he says that Cherry is now pregnant with his child. The survivors flee to Mexico, where they live in the ruins of a Mayan city.

Abernathy's (now ex-)boyfriend cheated on her with Daryl Hannah's stand-in - on her birthday. Oddly enough Zoe Bell, who plays herself alongside Abernathy (portrayed by Rosario Dawson), was Daryl Hannah's stunt double for both "Kil Bill" movies.

William and his colleague decide that self-preservation is the most important thing to think about with more infected sickos coming into the hospital, so William decides to 'get his wife'. Along the way, he looks into a bloody room and finds one of the infected killing a surgeon with a bone saw. He is soon attacked himself, but the power cord for the saw is pulled from the wall just in time (though it cuts his glasses in two). The sicko bursts an abscess on his face and smears it on Dr. Block. Meanwhile, Earl McGraw, the old Sheriff, is feeding his catatonic wife when he gets a call from Sheriff Hague about the incident. His wife soon turns into a sicko, so he is forced to kill her. Dakota escapes from her closet and jumps out a window to free herself from the hospital. When she gets to her car, she accidentally breaks her numb hand trying to open the door. Nonetheless, she gets home to find her son with the babysitter twins who are watching him and she sends them off. She gathers her son while he tries to gather his pets (a turtle, a tarantula, and a scorpion...in the same tank) and his pocket bike (which they place into the trunk) and they head to her father's place. When they get there though, she tells her son to stay in the car with the gun, and if any strangers come by, he's to shoot them. As soon as she walks to the door though, Tony shoots himself by accident. She rushes to the car to save him while the sickos ambush her, but her father, Earl McGraw, comes to her rescue and takes her and her dead son inside.

Stuntman Mike's next set of girls are in Lebanon, Tennessee, and they have their first encounter with him outside a convenience store. While Kim goes into the store for coffee and Red Bull, Abernathy sleeps with her feet out the window and Lee listens to her iPod. Stuntman Mike ends up sexually assaulting her (without her knowing) and he quickly drives off, watching the girls the entire time. When Abernathy goes into the store, she finds a magazine with Lee's picture inside and decides to purchase it. Lee is starring in a film nearby, while Abernathy and Kim are members of the crew. After leaving the store, they rush off to the airport to pick up their friend, stuntwoman Zoe Bell. They don't realize, however, that Stuntman Mike is there as well, taking pictures of the group of them.

She's actually named 'Jungle Julia' on her radio show. On her last show, she announced that she and her friends would be hitting the bars and that if a guy came up to her and bought her a drink, and then spoke a poem to one of her girlfriends (Butterfly), she would personally give them a lap dance. When Butterfly is later approached by Stuntman Mike, Julia says that she already gave up the lap dance. Mike sees right through this and Butterfly volunteers to the plan. The scene involving that event ends up being one of the missing reels.

Wray gets to the hospital and begins fighting off the infected to save Cherry who, losing her hopes of becoming a stand-up comedienne, is not happy that Wray finds jokes about cripples somewhat funnier. He attaches a table leg to her stump and they make their way to his truck. They both head off to The Bone Shack with the cops. Inside, they find JT unharmed having killed some of the infected in his restaurant. JT shows the group to a custom car and motorcycle that they can use to make an escape. In the meantime, they can rest in the safety of the restaurant. Cherry and Wray take the time to rekindle their relationship, and Wray shows Cherry the ring that he left for her in his jacket pocket, so they make love in the bedroom. The inscription gives Cherry a renewed sense of pride in her useless talents. At this point in the movie, the film cuts out. The next scene has the restaurant on fire. Dakota Block, Earl McGraw, the babysitter twins, and Cherry's old boss and go-go dancer colleagues have arrived, and Sheriff Hague has been attacked, however, he has new trust in El Wray, who tells him a convincing story and is now able to take all the guns. They decide to clear the way, make an escape route, and leave. Cherry decides to start up Wray's 'kill-dozer' and they take the car and motorcycle (while Ray takes Tony's pocket-bike) and they head to the Mexican border...but they're stopped by Lt. Muldoon and his troops.

After picking Zoe up from the airport, the girls head out for lunch discussing their recent flings, the movie they're working in, past stories, and the fact that Kim always carries a gun. Zoe soon brings up the fact that the one thing that she wanted to do when she travelled to America was get a muscle car, specifically a 1970 Dodge Challenger with a four-cylinder engine, and ride around in it to fulfill her dream. Luckily, she found one nearby. When she knew she was flying over, she did some research and found a car similar to the one from the movie "Vanishing Point". The girls decide to finish their meal and head directly to the farm where it's being held to take it for a 'test drive'.

After the night at the bar, the girls drive home down a dark road and Stuntman Mike follows closely behind in his stunt car. With the girls drunk and listening to loud music, he passes ahead of them, turns around, and strikes their car at a high speed, violently killing them all. Stuntman Mike is taken to the hospital for some small injuries (the most major of which is a broken thumb). The police have a feeling that he was responsible, but he is set free.

It turns out he doesn't really sell it; it's his own personal copy of "Italian Vogue". He sells it to Abernathy for 27 dollars. Abernathy wants to split it between her, Lee and another one of their girl friends. It is also at the Circle A parking lot that Stuntman Mike tickles Abernathy's feet and then pretends to have bumped into them while she was asleep.

Zoe talks to Kim about taking the car for a single ride in order to perform a stunt called the Ship's Mast, which they vowed they'd never do again after they did it in New Zealand. Kim finally gives in and decides to go along with it, but they need to con the owner into letting them drive the car in the first place. Abernathy realizes that they want to take the car without her, so she makes a deal that if she can get them the test drive, that she can come along with them. She ends up insinuating that Lee will sexually pleasure the man in exchange for a short test drive, and they head on their way, leaving Lee asleep and unknowing by the barn. While on the road, Zoe straps two belts to the hood of the car, hangs on, and lets Kim speed away down the road while she holds tight.

Quentin Tarantino is known for his "appreciation of feet" and often includes close-ups of female feet in his movies. In this movie, Abernathy gets her feet tickled by Stuntman Mike, Jungle Julia is barefoot on several occasions, and we see a close-up of Arlene's foot in a flip flop sandal. However, Pam's feet aren't seen throughout the movie.

While the survivors head to the top of the complex, Cherry uses her new leg to defeat the soldiers in their way. Abby tells them that they are all the antidote to the infection before attempting to lead the group through the troops. He is shot in the head on the way. Everyone else goes in the other direction. Underground, JT and Sheriff Hague set off the detonator that Wray gave to them, and they blow up the complex. They take that as their cue topside and Cherry is called into action. She single-handedly avoids catastrophe and destroys many of the containers of DC2 and fights off the troops. However, she is nearly shot, and Wray sacrifices himself to save her, getting shot in the process. While everyone boards the helicopter, Dakota investigates a second copter, finding her husband inside. She tells William she was seeing Tammy to hurt him, and before he attacks her with the red needle, her father saves her, shooting William multiple times. They board the copter, which begins to fly. Cherry's boss uses the copter's blades to decapitate the rest of the troops. Cherry says her final goodbyes to Wray, who she is forced to leave behind. He tells her that his child is inside of her, so she has to carry on and be strong. Dakota lets down a rope from the chopper and Cherry is pulled into the sky. The survivors make their way to Mexico, where there are more infected humans, but they hold their own inside the temples at Tulum. Here, with their back to the ocean, they are able to get by and repopulate the Earth. Cherry has Wray's child, and they survive day by day as 'two against the world'.
