All the choices are agents in one way or another. Pinky's nemesis is Professor Poofenplotz, first seen in "Isabella and the Tree of Sap". The other agents have nemeses that are not named.

For most episodes, Dr. Doofenshmirtz battles Perry the Platypus AKA "Agent P". Dr. Doofenshmirtz discovered his nemesis in the "It's About Time" episode.

In "No More Bunny Business", Perry the Platypus was battling a rogue agent, so Dr. Doofenshmirtz fought an agent of his own creation, Planty the Potted Plant. At the end of the episode, Major Monogram was accepting Planty into the agency. as he had defeated Dr. Doofenshmirtz.

Peter the Panda was introduced in "It's About Time" and fought Dr. Doofenshmirtz without Agent P knowing about it. Perry later finds out and nearly quits the agency. Later he and Peter combine forces with the other agents to defeat Dr. Doofenshmirtz and other evil scientists.
