1. Later on in the game, Batman is tracking one Talia al Ghul's personal assassins when he is ambushed by a small group of the assassins. Which of his allies comes to save him?

Answer: Robin

After being saved by Robin, Batman gives him a sample of his poisoned blood and instructs Robin to locate anyone with the poison in their veins. Earlier on in the game, the Joker poisoned Batman with his blood, and in order to coerce Batman into finding a cure, Joker sent out several other vials of his poisoned blood to hospitals in Gotham.

The poison is the after effects of Joker's Titan overdose, back in the first game "Batman Arkham Asylum".
2. Who is the second villain you encounter in the game, following your interrogation with Dr. Hugo Strange at the beginning?

Answer: Black Mask

As you break out of the interrogation chamber you are instructed to move into Line A. In the corner you can see Black Mask fending off several Tyger Guards with a chair. After the Black Mask you are confronted by the Penguin.

Answer: Alfred

After surviving his scrap with the Penguin and his thugs, Bruce Wayne manages to break into the City. He contacts his trustworthy butler Alfred, and requests he send a Batsuit into the city.

Answer: Two-Face

Two-Face is seething at Catwoman, as she stole from his safe, so in retaliation he is planning to execute her. "Fairly". Luckily Batman sweeps in and saves Catwoman, and restrains Two-Face.
5. Who does Batman encounter in the church, being held hostage by Harley Quinn, who also made an appearance in "Batman Arkham Asylum"?

Answer: Aaron Cash

Aaron Cash makes an appearance as a security guard at Arkham Asylum in the first game. He now works in Arkham City, at the Medical Centre in the abandoned church. Unfortunately he is later held hostage along with several other guards and doctors at the church by the villainous Harley Quinn. Mayor Quincy Sharp makes an appearance later on in the game, whereas Dr. Penelope Young and Frank Boles both perished during the events of the first game, both by the Joker.

Answer: The old GCPD forensics lab

Mr. Freeze used the old Gotham City Police Department Headquarters forensics lab to develop the cure, but was subsequently kidnapped by the Penguin, and put on display in Penguin's twisted museum. It seems that Mr. Freeze's cryogenics suit is also missing, and most likely claimed by Penguin.

"The Killing Joke" being the famous and iconic comic in which the Joker famously shoots and paralyses Barbara Gordon, ending her days as Batgirl. She decides to use her skills to good use however, and consequently becomes the Oracle.

Answer: The Riddler

The Riddler. Master of Puzzles. "Prince" of Enigmas. Who in "Batman Arkham City" kidnaps paramedics, doctors and security guards and tests Batman to overcome his elaborate puzzles in order to save them.

Answer: Nightwing

Nightwing is a playable character in the challenge maps, and doesn't make an appearance in the games main storyline. He along with two additional challenge maps are a part of a DLC for the game.

Answer: Ra's al Ghul

Ra's al Ghul was the real mind behind Arkham City, and used Strange as his puppet. When Strange failed to stop Batman, Ra's stabbed Strange in the back. Literally.
