In the first half of the game, Wheatley provids Chell (the protagonist) with assistance in trying to escape from the Aperture Science facility. In the second half (SPOILER ALERT), he turns on Chell, forcing her deeper into the bowels of the compound and forcing Chell to make her way back to him.
