Benoist was previously one of the cast members of "Glee". She also has guest appearances on "Flash" and "Legends of Tomorrow" as the character.

Henshaw was the leader of the DEO, but had died in an accident in Peru years before the pilot episode. The Manhunter assumed his identity at that time.

Schott is Supergirl's best friend in the pilot, and a computer expert at CatCo. in season one. In season two he took a job working directly with the DEO. His father is the villain known as the Toyman, that was addressed in a season one episode.

Gustin stars on "The Flash" television series as the titular character. He's appeared in several crossover episodes of "Supergirl" and also on "Legends of Tomorrow" and "Arrow" as the same character. Barry Allen is one of several characters from those various series to crossover on the other series'. Gustin was also a cast member on "Glee" against Melissa Benoist before getting his own series.
