"Dead Man's Blood" involved vampires that killed Daniel Elkins and stole the Colt revolver. Vampires were also key to "Fresh Blood". In that episode, Gordon attempted to kill the Winchesters as they hunted a vampire. The vampire captured Gordon and turned him into a vampire. Sam ended up killing Gordon.

The brothers faced an arachne after it lured Sam to Bristol, Rhode Island to get revenge for Sam's actions during his time without a soul. Sam had used the human sheriff as bait. The sheriff was turned into an arachne and then, since Sam did not realize what had happened, he shot the sheriff thinking it was a merciful death.

Instead, the sheriff had become an arachne that survived and turned several women into creatures as well.
