Crowley's snarky reference was to Sam's actions during the year Dean was in Purgatory, when, overcome with grief, Sam ran until he hit a dog and ended up in a relationship with the veterinarian who saved the dog. The Mark of Cain allowed Dean to overcome the spell. He then stopped his brother from killing himself by using his own blood. Blood from both brothers was then enough to open the box. After, Dean told Sam, "That says something, doesn't it? Werther splits us up in there, and within an hour, we're both on the brink of death? . . .

The universe is trying to tell us something we both should already know: We're stronger together than apart."

Ironically, immediately after Dean killed Death to save Sam, Sam's plan to remove the mark was implemented by Rowena, Castiel and Crowley, and the Darkness was released.
