Shortly after Dean said this in "Family Matters", the alpha vampire escaped from the cage. Later in the episode, the brothers discovered that Samuel was working for Crowley.

This scene from "Clap Your Hands if You Believe" was very funny, first because soulless Sam was eloquent in his disdain and then because Dean had to struggle to explain empathy to him, and Dean was forced into a role reversal and had to be the nice guy dealing with people. The irony, of course, was that the woman was right about what creature the brothers were hunting.

In "Family Matters", Crowley said this to the brothers after claiming he was the one that could get Sam's soul back and threatening to put Sam back in Lucifer's cage if the brothers did not work for him, bringing him creatures. This reference to "Charlie's Angels" was just one reference to a classic television show in the episode.

At another point, when Dean met Christian in the Campbell compound, he said, "Hello, Newman" with the same inflection Jerry used when he'd meet Newman in "Seinfeld".

This confrontation between Castiel and Crowley in "The Man Who Would Be King" had several levels. First, it was funny. Second, Crowley sort of ran through the brothers' greatest hits. Third, since Castiel put the Winchesters off limits, it set up the next episode when Crowley went after Lisa and Ben. And, finally, it showed how desperate Castiel was becoming.
