The chest only appears if you witnessed the scene where the father of the unwed man is putting the chest back there. This can be seen during the Commsphere Network part of Chapter 4. You later have the option to keep the gil or return it.

The Besaid Key is used to open the chest in the Temple. The chest contains a search sphere, whose purpose is to reveal the four other ciphers on the island, which open the wall passage in the cave on Besaid. At the end of the path behind the wall, there is a garment grid!
