Manco is the Spanish word for someone who has a lame hand. The Italian equivalent is 'Monco', and is the spelling usually listed in sources about the movie. Manco somewhat lived up to this name by using his left hand for most things, which gave the appearance that he was indeed 'manco'. He mostly only used his right hand to draw his pistol.

In the movie, Manco was a bounty killer. He most always wore a brown poncho with white designs and was smoking a cigar.

He told him that he couldn't just stop the train like that, but when he saw Mortimer's gun, he changed his opinion and got back on the train.

Indio was the leader of the gang that planned to rob the Bank of El Paso. He had curly hair and a beard, and he laughed a lot. His "Wanted" poster even had a picture of him laughing on it. Mortimer to him. Manco also paid him to tell him where Mortimer was staying. Later, the boy also told him about some of the gang members coming to town in exchange for more money from Manco. Whenever Fernando had something to tell Manco, he would stand outside the hotel and whistle to get Manco's attention.

Like the other children in the town, Fernando wore white clothing and was often seen running around with the other kids. He was also hidden under a porch during the showdown between Mortimer and Manco.

Manco and the three men were in the desert before they were supposed to rob the bank in Santa Cruz. He said that he could do so without blowing it up and ruining some of the bank notes in the process. He then used a drill and some acid to help pick the lock and open the safe.

Before becoming a bounty killer, Colonel Mortimer had actually been a soldier. According to the old man that Manco got information about Mortimer from, Mortimer had once been "the finest shot in the Carolinas". He was always wearing black and was almost always smoking a pipe.

Indio carried a gold pocket watch with a woman's picture in it. Whenever he was about to shoot someone, he would open the pocket watch and let its music play. He would tell them that they needed to draw their weapons once the music stopped and try and kill him.

He would also sit with the pocket watch open and listen to the music while he had flashbacks of a crime he had committed where he killed a man and raped his fiance. The woman he was raping killed herself during the act with a gun that had been under the pillow, leaving Indio somewhat traumatized.

After Mortimer shot Groggy, Indio shot Mortimer's gun out of his hand. He then opened his pocket watch to let it play its music. He told Mortimer to pick up his gun and try and kill him when the music ended. While the music played, Mortimer's eyes looked tear-filled. Before the music stopped, Manco showed up with Mortimer's pocket watch which was nearly identical to Indio's and played the same music. While Manco pointed his gun at Indio, he handed his gun belt to Mortimer. Once the music from Mortimer's watch stopped playing, Mortimer shot Indio and picked up his pocket watch.

When Manco commented on the family resemblance between Mortimer and the woman in the pictures from both watches, Mortimer revealed that the woman that Indio had raped was his sister. Mortimer then told Manco to keep the entire reward because he deserved it. This showed that Col. Mortimer's true motive for following Indio was vengeance for his dead sister rather than the reward.
