This is Jesus' response to Satan following one of Satan's temptations. Jesus had been alone in the desert for forty days after his baptism before Satan had arrived. In this instance, Satan had taken Jesus to the top of a very high mountain, shown him all the kingdoms, and said "All this I will give you if you will bow down and worship me."

Nicodemus was confused by this, so Jesus had to explain that this means being born of the water and the spirit.
