Elaine Stritch portrayed Colleen Donaghy, the caustic, overbearing mother to Alec Baldwin's Jack Donaghy. Jack got so riled up by his mother during her visit that he suffered a massive heart-attack. "Conflict is great acting material," said Elaine about her relationship with her television son. "You show me two people that are getting along and having a cup of tea, I don't want to watch that on television." Elaine Stritch is best known for originating the role of Joanne in "Company" on Broadway, and for her one-woman show "Elaine Stritch: At Liberty".

Isabella Rossellini appeared in two episodes of the first season of "30 Rock" as Jack's ex-wife, Bianca. She got into a vicious catfight with Liz when she was led to believe that she was dating Jack, and showed up again in the following episode to finalize her divorce. Isabella did appear as herself in another NBC comedy, "Friends", in the episode where Ross removed her name from his "freebie list", only to bump into her in the coffee shop. Both Conan O'Brien and Al Gore have appeared on "30 Rock" as themselves, and in "The Rural Juror", Whoopi Goldberg was the celebrity who stepped in to endorse a "meat machine" in the Ukraine after Tracy pulled his support for the defective product.

Prince Gerhardt was portrayed as a grotesque cripple with a lame hand and a body that couldn't produce joint fluid. "Most people in his situation would be angry with his family for the centuries of inbreeding, but not Gerhardt," explained Jack. "He's too busy trying to stave off infection." Jenna crashed the party in the hopes of rubbing shoulders with royalty, but it all ended badly when Gerhardt died from an allergic reaction to champagne. Best known for creating and portraying the Pee-wee Herman character that appeared in movies and television shows during the 1980s, Paul Reubens was the perfect choice to play the disease-addled Prince.

His appearance on "30 Rock" was disturbing, short-lived and utterly hilarious.

Sean Hayes is best known for his role as the flamboyant Jack McFarland in "Will and Grace", but on "30 Rock" he played a character that was the polar opposite of Jack. Jesse Parcell was Kenneth's hillbilly cousin who kept talking about wolves, and was so star truck with Tracy that he held him against his will when he wanted to return to New York.

The casting of Sean Hayes was oddly fitting, because Jack McBrayer (who plays Kenneth) has been mistaken for Sean in the past by autograph hounds, who refused to believe that he wasn't the "Will and Grace" star. "I didn't want to get him in trouble," said McBrayer, "so I'd sign 'Sean Hayes' and go about my day."

In the first episode of season two, Jack had his tech guys digitally capture Jerry Seinfeld from the hundreds of hours of footage of "Seinfeld" owned by NBC so that he could be inserted into the current crop of programs airing on the network. Jack coined the process "Seinfeldvision", which saw Jerry Seinfeld inserted into episodes of "Law and Order", "Heroes", "Deal or No Deal" and the fictional reality show "MILF Island" without his consent. Ironically, Jerry's appearance on "30 Rock" occurred a few weeks before the release of the animated film "Bee Movie", which Jerry had starred in and co-written, and a few seconds of the trailer for that movie could be seen on Jack's laptop during the episode.

In the first two seasons of the show, Jack was constantly competing for advancement within General Electric with Devon Banks, portrayed by Will Arnett, the actor best known for playing Gob (pronounced "Job") on "Arrested Development". For his appearances in the show's second season, Arnett was nominated for an Emmy for Outstanding Guest Actor for a Comedy Series, as were Steve Buscemi and Rip Torn, but they all lost to Tim Conway, who won the award for playing Bucky Bright, the former child star who regaled Kenneth with hilariously inappropriate stories about the good old days in the episode "Subway Hero".

When NBC announced a new company-wide global eco-initiative to save the Earth so that they could "drain the remainder of its resources", Jack found a way to capitalize on the environmentalism trend by introducing "Greenzo", an environmental advocate whose motto was "Saving the Earth while maintaining profitability". Greenzo was portrayed by David Schwimmer in a green and yellow costume and cape.

After success got to Greenzo's head, Jack was forced to fire him but he still needed a spokesperson, so he lured Al Gore to NBC by telling him that General Electric had perfected a trash-powered car.

Jason Sudeikis had a recurring guest role on "30 Rock" as Liz's boyfriend, Floyd. Jason has been a repertory player on "Saturday Night Live" since 2006, and had previously worked with Tina Fey when she was the head writer and a fellow performer on "SNL".

He is best known on "SNL" for his impersonations of Joe Biden and "American Idol" winner Taylor Hicks. The other choices were all current or former cast members on "SNL" who also guest starred on "30 Rock" - Chris Parnell has a recurring role on "30 Rock" as Dr. Spaceman; in "Somebody to Love", Fred Armisen played Raheem Haddad, the Middle-Eastern neighbor that Liz suspected was a terrorist when he was just a fan of "The Amazing Race"; and Will Forte had a small role as Prince Gerhardt's page in "Black Tie".

Edie Falco made her mark in various supporting roles in film and television before hitting it big with her portrayal of Carmela Soprano on "The Sopranos". The role earned her numerous Emmy, Golden Globe and Screen Actors Guild Awards, and she became the first actress to win all three accolades in the same year for playing the same role. For her comedic turn on "30 Rock", Edie was nominated for an Emmy for Outstanding Guest Actress for a Comedy Series, but lost to Kathryn Joosten, who played Karen McCluskey on "Desperate Housewives".

Both Nathan Lane and Matthew Broderick have guest starred on the show. In the first season episode "The Fighting Irish", Nathan Lane played Eddie Donaghy, Jack's estranged brother who reconciled with the rest of the Donaghy children over the apparent death of their father, only to be revealed in the end of the episode to be running a scam.

In the season two finale "Cooter", Jack left General Electric and was appointed Homeland Security Director for Crisis and Weather Management, a portfolio that involved "homeland security, extreme weather preparedness and the war on the poor". Matthew Broderick played James Riley, Jack's colleague in the Government who was given the nickname "Cooter Burger" by President Bush because he looked like a turtle, and was once spotted eating a sandwich (which the President mistook for a burger).
