Answer: The Colt revolver

Dean killed Azazel by shooting him with the Colt revolver in "All Hell Breaks Loose, Part 2". Azazel was about to kill Dean when John climbed out of Hell and grabbed the demon, distracting him long enough for Dean to retrieve the gun and kill Azazel.

Answer: Holy oil

Castiel and Dean used a burning circle of holy oil to trap Raphael in "Free to be You and Me". The holy oil was used many more times, including its use against Gabriel in "Changing Channels" and against Castiel in "The Man Who Would be King".

Answer: Silver knife dipped in lamb's blood

In "What Is and What Should Never Be", Dean killed a djinn using a silver knife dipped in lamb's blood. The djinn was draining him before he started pulling himself out of the fantasy state the djinn had created. Sam found Dean and helped him, and then was attacked by the djinn himself. Dean killed it, saving his brother and himself.

Answer: Machete

While dead man's blood was used to make vampires sick, the Winchesters had to chop off their heads to kill them. John killed a vampire with a machete in "Dead Man's Blood" and Dean killed many vampires that way in "Live Free or Twihard". Sam also killed the vampire Gordon by cutting off his head with razor wire, and John used the Colt revolver to kill one in "Dead Man's Blood".

Answer: Knife dipped in Dean's blood

In "Sex and Violence", the siren poisoned Sam and Dean with its saliva. Bobby stabbed Dean to get his blood on the knife and then threw the knife into the siren to kill it and save the Winchesters.

Answer: Borax

The Winchesters and Bobby tried all the weapons they knew of to harm the leviathans, but it was not until Jody accidentally spilled some cleaner containing borax at Bobby's that they discovered something that hurt the creatures. Bobby also discovered that you could chop their heads off - although the head would reattach itself eventually.

These two discoveries occurred in "Slash Fiction".

Answer: Iron ax

Sam used an iron ax to kill Leshi in "Fallen Idols". The god was about to kill Dean. Dean told the god that it could not kill him, since it only killed its fans, and the god was in the form of Paris Hilton. A funny inside joke occurred when Dean said he had not even seen "House of Wax".

This was a movie Paris Hilton was in, along with Jared Padalecki. Leshi then went after the ax, since it had belonged to Dean's father, and Leshi could take John Winchester's form to kill Dean. Before Leshi succeeded, though, Sam killed the god using the ax.

Answer: Burning the remains

While they occasionally found another way to help a ghost move on, such as helping Constance face her children in "Pilot" or talking to Molly so she could let go in "Roadkill", the vast majority of ghosts were dispatched by digging up and burning their remains, or burning some other remains, such as hair.

In "Time is on my Side", the brothers realized that Doc Benton could not be killed. In order to stop him, the brothers locked him in a coffin and buried him. Sam, desperate to save his brother, argued that Dean should consider using the doctor's formula to make himself immortal and save himself from Hell, but Dean refused.

In "All Dogs Go to Heaven", the brothers used silver bullets, shooting the creatures in the heart. Skinwalkers were described as "werewolf cousins" and were killed the same way.

The brothers used a flamethrower to kill the changeling in "The Kids are All Right". After killing the changeling mother, the changeling children burned up, as well. This was the first episode featuring Lisa and Ben, who were recurring characters in season 6.

The god was about to have Dean killed in the episode, and Sam saved him. Sam did use a ram's horn, but the weapon only put the god out of commission for a couple centuries. When Bobby could not find a way to kill the god for good, he told Sam to do this, telling him that the next time it turned up, it would be dealt with by a "hunter in a spacesuit."

The brothers killed numerous demons, including Azazel in "All Hell Breaks Loose Part 2" and his son Tom in "Devil's Trap", with the Colt revolver. They also used Ruby's knife to kill a lot of demons. And, Sam used the power he got from the demon blood to kill demons, including Azazel in "On the Head of a Pin" and Lilith in "Lucifer Rising".

While the brothers performed a lot of exorcisms, that weapon simply sent the demons back to Hell, but did not kill them permanently. For instance, they exorcised Meg in "Devil's Trap", but she returned to possess Sam in "Born Under a Bad Sign".

Answer: Brass pipe

In "Everbody Loves a Clown", the brothers needed a brass knife to kill the rakshasa. When they could not find one, they went to the funhouse to get a pipe. While the invisible creature threw knives at them, Sam was able to get the pipe and Dean opened a vent to release steam so he would see the creature.

He was able to get a glimpse of the figure behind Sam, and Sam stabbed and killed it.

Answer: They did not kill him

The brothers tried to kill Lucifer using the Colt revolver in "Abandon All Hope", but discovered it would not kill him. In "Hammer of the Gods", Gabriel told them, via videotape, that they could use the rings from the Four Horseman to open the door to Lucifer's cage and lock him back in.

The brothers got the rings and then Sam let Lucifer possess him so he could jump in the cage. It did not kill the devil, but locked him away back in Hell. That scene occurred at the end of "Swan Song".
