The setting for the film is San Francisco. This location was chosen mainly to provide a spoof replica of the car chase sequence in the 1968 film 'Bullitt' which was also filmed in the city.

There are in fact four plaid overnight cases. The first bag belongs to Howard Bannister and his case contains various igneous rocks. The second case is owned by Judy Maxwell and she has in it just her clothes and a dictionary. Mrs Van Hoskins, a wealthy older lady, owns the third case in which she stores her expensive jewelry.

The fourth case has in it top-secret government papers and the bag belongs to the mysterious Mr Smith.

Judy decides to play games with him and before she leaves the store picks up a radio and puts it on the counter where she tells the cashier that Howard will pay for it.
The San Francisco Hilton Hotel was used as the location for the lobby and pharmacy sequences of the fictional 'Hotel Bristol.' The sequences in these parts of the hotel were shot late at night when the area required for filming could be closed off.

The background music played during the American Musicologists banquet was taken from Thoinot Arbeau's "Orchesographie", which was a study of the late sixteenth century French renaissance social dance.

The pieces were unusually played in this instance on a Hammond organ and a sitar.

Eunice is suspicious and does not believe Howard when he says that it is the television programme that she can hear in the background. Eunice tells Howard she is going to come to Howard's room at once so Howard tells Judy she has to hide and pushes her onto the ledge outside his hotel room window.
When Judy is on the ledge of the hotel outside Howard Bannister's room, the shots were actually filmed on a sound stage in Burbank. The long shot of Judy actually hanging from the ledge was performed by a stunt woman and not Streisand, the location for this shot being at an unnamed building in Westwood, California.
'Whats Up Doc?' was the first American film to show the names of the stunt actors in the rolling credits.

The song 'As Time Goes By' was recorded live by Streisand at the time the scene was shot so that a natural sound would be reflected as she stepped down from the top of the piano.

Howard has already discovered that he has the case with the jewels and and that Judy has the case with the Top Secret documents.

While fighting breaks out Judy and Howard grab all four plaid cases and make their getaway on a grocery delivery bike parked outside. Ryan O'Neal's brother Kevin played the part of the delivery boy whose bike is taken by Howard and Judy.

The scene in the court follows the frantic car chase through the streets of
San Francisco which had ended up by he Sausilito Ferry dock. Howard and Judy had attempted to reach the departing ferry before the doors close but miss and they and a couple of the cars following in the chase end up floating in the bay. All parties involved in the chase and the arresting police officers end up at the court house where the judge asks for an explanation as to what has been going on. After several aborted attempts to obtain the information the judge asks the the 'quiet one under the blanket' to give an account of events. as Judy removes the blanket from her head she looks up at the judge and says "Hello, Daddy."
The chase scene took 19 days to shoot, using 32 stunt men and cost one million dollars. The scene lasts for a total of 11 minutes of the film's duration.
