Jessica Heafey played Gwen Campbell, Corin Nemec played Christian Campbell, and Davd Paetkau played Mark Campbell. They were all distant cousins of the Winchester brothers, on their mother's side. Mark was killed in "Exile on Main Street". It was revealed that Christian had been possessed by a demon in "Family Matters" and the demon and Christian were later killed in "Caged Heat". Gwen was killed in "And Then There Were None". Sonya Salomaa played Rachel, an angel that reported to Castiel. She was killed in "Frontierland".

Julian Richings played Death. He retrieved Sam's soul from Lucifer's cage in "Appointment in Samarra". He also put up a wall in Sam's head to protect him from the memories of Hell. Castiel later destroyed the wall at the end of season 6 in "The Man Who Knew Too Much".

Mitch Pileggi played Samuel Campbell, Mary's father and the grandfather to Sam and Dean. Samuel died in 1973, and Crowley resurrected him to help him find Purgatory. Samuel lied to his grandsons about his connection to Crowley until it was revealed during "Family Matters".

He also betrayed them during "Caged Heat". Samuel told the brothers he was working with Crowley so Crowley would resurrect Mary. Samuel was killed for the second time during "And Then There Were None".

Sebastian Roche played Balthazar, the angel that stole the weapons of Heaven, as revealed in "The Third Man", and unsank the Titanic, as revealed in "My Heart Will Go On". He also provided Sam the spell to try to keep his soul out of his body, which included killing Bobby, in "Appointment in Samarra", transported the brothers into an alternate reality in "The French Mistake", and later assisted the Winchesters in trying to stop Castiel from opening Purgatory in "The Man Who Knew Too Much".

Jim Beaver played Bobby Singer. Bobby was possessed by the worm monster in "And Then There Were None", and he stabbed and killed his friend Rufus Turner. The worm also caused Dean to kill Gwen, and was in Samuel when Sam was forced to kill him.

Jensen Ackles played Dean Winchester. It was revealed in "Exile on Main Street" that he had lived with Lisa and Ben for a year. He was also poisoned by a djinn in "Exile on Main Street", turned into a vampire in "Live Free or Twihard", abducted by fairies in "Clap Your Hands if you Believe". and wore Death's ring for a day in "Appointment in Samarra". Through this all, he continued to hunt supernatural creatures and save people.

Kim Johnston Ulrich played Dr. Eleanor Visyak. She first appeared in "Like a Virgin", when she gave Dean a sword to stop the dragons. It was later revealed in "Let it Bleed" that she was actually a creature from Purgatory that had escaped after the author H.P. Lovecraft opened a door to that dimension.

The character died from wounds inflicted by Crowley and Castiel in "The Man Who Knew Too Much".

Mark Sheppard played Crowley, the King of Hell. Bobby discovered that Crowley's real name was Fergus MacLeod, and he was from Scotland. In "Weekend at Bobby's", Sam and Dean went to Scotland to dig up the demon's bones and force him to give up his claim on Bobby's soul.

Lanette Ware played Raphael, the archangel engaged in a civil war in heaven. Raphael was leading those that wanted to go back and play out the Apocalypse, while Castiel was leading those that wanted to stop them. Raphael appeared in "The French Mistake" and "The Man Who Knew Too Much". Castiel killed the archangel in the latter episode.

Jared Padalecki played Sam Winchester. It was revealed during season 6 that Sam's soul was tortured in Hell for 18 months, which is 180 years in Hell time. He was separated from his soul during that time, and then protected from those memories while a wall was in his brain. Once the wall was removed in "The Man Who Knew Too Much", his psyche split into three parts and he had to reassemble the parts.

The character also killed his grandfather in "And Then There Were None", while his grandfather was possessed by a monster. Through it all, he continued to hunt supernatural creatures and save people.

Julia Maxwell played Eve. She first appeared in "Like a Virgin" when the dragons used a virgin sacrifice (also played by Julia Maxwell) and a spell to open a door to Purgatory and let her out. Eve, the "mother of all" monsters, proceeded to rally all monsters and create more monsters to try to destroy mankind.

She was finally killed by Dean in "Mommy Dearest", after he swallowed phoenix ash (deadly to her) and taunted her into biting him.

Rachel Miner played Meg. She told Sam and Dean this during "Caged Heat" to convince them that she would be able to force Crowley to give them what they wanted. The brothers worked with her to try to force Crowley to give Sam his soul back. Crowley admitted he could not retrieve it though, and Meg got away before the brothers could kill her.

In "Two and a Half Men", the Alpha Shapeshifter stormed the Campbell compound to retrieve the shapeshifter baby. When he first arrived, he took the form of Samuel, played by Mitch Pileggi. He then went to the panic room, in the form of Sam, played by Jared Padalecki. Finally, he morphed into Dean, played by Jensen Ackles, before wresting the baby out of Dean's arms.

The Alpha Shapeshifter was also briefly played by Mark Sheppard, who plays Crowley. Mark Sheppard played the Alpha in "Caged Heat" when Crowley was torturing the Alpha while the Alpha was in his form.

Rick Worthy played the Alpha Vampire. During "Family Matters", he told the brothers that the reason Samuel was kidnapping and torturing creatures was to find Purgatory. He also told them that Samuel was working for someone else. Soon thereafter in the same episode, the brothers learned that it was Crowley calling the shots.

Misha Collins played Castiel. On the pretext of talking to God, Castiel narrated this episode, explaining how he partnered with the King of Hell in order to win the war against the more powerful Raphael. He was seeking guidance in this episode, which he did not get, and therefore continued on his path over the next two episodes, killing Balthazar, breaking the wall in Sam's head, opening the door to Purgatory, and consuming all the souls within.
