Miss Scarlet, Colonel Mustard, Mrs. White, Reverend (or Mr.) Green, Mrs. Peacock and Professor Plum are the six original clue suspects who have appeared in almost every iteration of the game. They are suspects in the murder of Mr. Boddy (or Dr. Black). Typically speaking, the characters would mostly have the same personalities from game to game but there might have different backstories. Miss Scarlet is the typical femme fatale. Colonel Mustard is an old English military man. Mrs. White is a housekeeper. Rev. Green is a shady vicar. Mrs. Peacock is a middle-aged wealthy widow. Professor Plum is a research professor usually studying Egyptology or archeology in some form.

Later editions of "Clue" sometimes expanded the cast of suspects. The VCR Game adds Madame Rose, who is Mr. Boddy's sister and into the occult, Miss Peach, a young American who is usually a long-lost relative of Mr. Boddy and Monsieur Brunette, a con artist.

In 2002, Hasbro released a few "Clue" games with interconnected storylines. "Clue FX" adds two new suspects, Mrs. Meadow-Brook, whose husband is Mr. Boddy's lawyer and the game's victim, and Rusty, the gardener. Lady Lavender, Prince Azure and Lord Grey (in addition to Miss Peach) are non-suspect player characters although they all become suspects in other games in that series. Lady Lavender is an herbalist. Lord Grey (who appears to be a totally different character from previous iterations of people named Grey) is a shady architect and Prince Azure is wealthy art dealer who also sells black market weapons.

In 2016, Dr. Orchid, a botanist, was added to the remake and replaced Mrs. White. These are just some of the new and unusual characters added to the game during the years. Typically, the additional characters are not permanent fixtures but tend to enter and leave the game periodically and depending on the edition.
