Nancy was given a recommendation by Franklin Rose to jobshadow archaeologist Jon Boyle. Jon is the expedition leader of a dig about 100 kilometres from Cairo. It is the tomb of a lost queen of Egypt. Franklin Rose first appeared in the sixth game in the series "Secret of the Scarlet Hand", as a board member of the Beech Hill museum in Washington. In the opening cutscene there is a flashback of an expedition 60 years ago to the tomb. The expedition members disappeared and were never found. The game then switches to present day, where a sandstorm appears in the camp. After the storm dies down, Jon is found lying on the ground. He later calls Nancy from the hospital, explaining to her that someone knocked him out.

Nancy lives with her father Carson Drew in the city of River Heights. Her mother died at an early age. The family is assisted by housekeeper Hannah Gruen. The last inhabitant of the house is dog Toto. The location of River Heights was never specified in the Nancy Drew books. It is generally considered to be somewhere in the midwestern part of the United States. The city is near Bayport, where Frank and Joe Hardy live. Frank and Joe are also detectives. Nancy has traveled to New York in game two of the series "Stay Tuned For Danger", where she tries to solve the mystery of someone threatening a soap star. In game three of the series she visited San Francisco. In this game called "Message In A Haunted Mansion", she tries to find out who is sabotaging the renovation of a mansion. In this game "Alibi In Ashes" Nancy enters the annual Clue Challenge in her home town. Teams have to follow cryptic clues in order to find a specific location. At that location they will find a medal. The winners will be able to put something of their choice in the time capsule to be buried at the end of the challenge. The unique premise of this game is that you will not only play as Nancy (like in the other games), but can also switch to her friends Bess, George and Ned to perform specific tasks within the game.

Nancy has been asked to investigate a series of monster sightings at the German Castle Finster by its owner, Markus Boehm. Apparently the monster has been spotted near the castle grounds for centuries, returning to the woods again after finding a new victim. The opening sequence shows the player a scene from the past where a young girl picking herbs is chased by a monster. When she arrives at the castle gate, she finds it closed and is then dragged off by the monster. The scene then moves to the present day with Nancy arriving at the castle gate. She also finds the gate closed (luckily no monster drags her off). When she rings the bell, someone called Lukas calls out to her and refuses to let her in, unless she passes a test. A bucket is lowered containing a game where Nancy has to match two monsters that are the same from a row of monsters. After passing this test, Lukas wants to give her another test, but fails to do so, as the mayor of the castle community, Karl Weschler, arrives and chases him away. Nancy later learns that Lukas is the son of the head of security at the castle and a very mischievous child (Karl refers to him as the castle's other monster). He is very fond of pranks, usually targeting Karl (since he is so good at noticing). Nancy can usually find him in the lobby, where he challenges her to a game called "Monster". In this game the two players play either the farmer or the monster. The players that is the monster hides three of them in a herd of cows. The object for that player is to kill as many cows as possible, while the farmer tries to locate the monsters as soon as possible (by looking at which cows move after a cow was killed). In the second round the roles reverse.

In "Tomb of the Lost Queen" Nancy goes to an archaelogical dig near Cairo. At this site might be the tomb of one of the lost queens of Egypt, Nefertari. Nancy is there to job-shadow Professor Jon Boyle, the leader of the dig. Also working on the dig is Dr. Abdullah Bakhoum, an eminent Egyptologist. His knowledge of Egyptian history and hieroglyphs is as large as his ego is, so Nancy finds. He holds very strong opinions about various issues and talks in a rude and condescending manner about those things he does not appreciate. He is sure the tomb is that of Nefertari, as he does not come out of bed for anything less than spectacular. Abdullah has taken on archaelogy student Lily Crewe as his assistant. Professor Boyle tells Nancy that it would not have been his choice given her history, but refuses to say more on the issue. During the course of the game Nancy unearths that Lily was involved in a plagiarism scandal. Yet Abdullah has given her a second chance. As it turns out, Abdullah is pressurizing Lily to do his dirty work for him. Abdullah has ties with the black market and is behind the attack on Professor Boyle, who had to go to a hospital in Cairo as a result. He also arranges several other accidents to happen, mostly through Lily. In the end sequence Nancy has to find a path to the exit of the tomb to prevent Abdullah from escaping. When she arrives at the exit, Abdullah tries to attack her, but is struck down himself by Professor Boyle, who arrived back at the dig just in time to save Nancy.

Rentaro Aihara is the handyman at the Ryokan Hiei in Kyoto, Japan. Nancy was invited to stay at the ryokan as a gift from P.G. Krolmeister for solving the crime in the previous PC game "Trail of the Twister". The ryokan is allegedly haunted and has a reputation among ghost hunters. Rentaro has lived next door to the ryokan, since age seven. He is a friend of the Shimizu family, who own the ryokan. He spends most of his time in his shed in the garden, where he is working on a robotic dog. He has previously created a robotic cat for his girlfriend Miwako Shimizu. It is Rentaro's dream to move to the city and get a job at a technology firm. Miwako does not want to leave the ryokan however. When he fails to convince Miwako, he sets up various hauntings at the ryokan, scaring guests away. That way Miwako might finally want to leave to the city with him. Naturally Nancy eventually foils his plot and he leaves the ryokan and loses the love of Miwako. Nancy first meets Rentaro near the public baths at the ryokan. Rentaro explains to her that the colour of the curtain hanging at the entrance of the baths indicates whether the bath is for men or women only that day. Blue means men only and red means the bath can be used only by women that day.
