Departing from Pinang Peranakan Mansion in George Town, Malaysia in the morning, teams were told to take a taxi to the bus station, a bus to Kuala Lampur's train station, and a train to the country of Singapore to locate their next destination, the Victoria Concert Hall. Once there, teams needed to receive a clue from "The Amazing Race Asia" host, Allan Wu. One of the clues received was the race's only Fast Forward. Only Jordan and Jeff and Monica and Shawne completed this task. In 'Horse Power', everyone else played a quick game of polo on a wooden 'practice pony'. Arriving in Santiago, the capital city of Chile, teams made their way to the bus stop to reach Chile's second largest city, Valparaíso, where they could find their first clue box of the race. Arriving at Terminal San Borja in Santiago, many teams boarded the first bus together, but some were forced to take later buses departing only minutes later. One team, Jet and Cord, had the wrong currency altogether and needed to exchange it elsewhere. Everyone arrived in Valparaíso not long after.

Both tasks required that the message be attached to a carrier pigeon and sent to a headquarters.

Louie and Michael, the first to arrive at the U-Turn, decided to use it to make Joe and Heidi, only moments behind them, finish the 'In the Trenches' task. This ultimately led to their elimination.

After a lengthy leg in Shanghai, Louie and Michael ended up stepping on the mat after sunset and they became the final team eliminated from the race.

At Sentosa Island, teams took a steep zipline twelve hundred feet through a jungle canopy to get their final clue sending them to the Pit Stop at Singapore's Marina Barrage. Dan and Jordan, having won the race's only Fast Forward task, ended up taking their first first place victory of the race. They both won motorcycles, of all things. Carol and Brandy, U-Turned earlier in the leg, were almost able to catch up but failed to do so in the very end and became the seventh eliminated team.

When teams landed on the shore, they opened their bottles to find a map leading to their next Pit Stop, the Paradise Sun Hotel down the beach. Steve and Allie were first to arrive (though they left all of their bags behind at the Detour and would not be able to claim them). For their victory they both won $7,000 (each) as well as dinner on the beach, massages, and as much 7-Up as they could drink (courtesy of 7-Up, this being the seventh leg of the race). After tons of mistakes by several teams in this leg, it was Jet and Cord who arrived at the mat in last place. The two of them were spared by a non-elimination leg, though.
