Pompe Disease is a metabolic disorder that disables the heart and skeletal muscle. It is very rare (1 in 40,000 births) and up until very recently was a fatal disease. The disease is caused by a genetic mutation in a gene that makes an enzyme used to break down Glycogen. Because the body cannot break down Glycogen effectively there is a significant increase in cardiac muscle and the heart enlarges. Usually this enlargement takes up the entire chest cavity and typical symptoms include muscle failure, inability to speak or swallow, severe weakness, and ultimately breathing problems.

Megan, the oldest of the two Crowley children, meets Dr. Stonehill at the door and has a conversation where she asks him about his hobbies. Later in the movie, John Crowley visits the doctor at his home to have a meeting. Stonehill throws paperwork at John then leaves to go bass fishing.

John Crawley arrives home to tell his wife that he has basically promised Dr. Stonehill half a million dollars from his fictitious, and yet to be created foundation. John and his wife Aileen immediately start fundraising with friends, relatives, and through special events. When Dr. Stonehill comes to visit they present him with a down-payment of $92,000 to get started.

Dr. Bob Stonehill is a researcher at the University of Nebraska in Lincoln. In reality, Dr. Stonehill does not exist. The enzyme cure for Pompe Disease was developed by Dr. Yuan-Tsong Chen. At the time, Dr. Chen was a researcher, professor and chief of medical genetics at Duke University Medical Center.

The family are altogether on an outing to the lake. There is a scene where the children are in their wheelchairs throwing bread crumbs to the ducks. Patrick has the bread in his hand but suddenly cannot move his hand. His dad makes him smile by helping him throw the bread. Later in the film, Megan is at the fair on her birthday and has the same inability to toss a ball in a fairground game.

John Jr. is the oldest of the three Crowley children. In one especially touching moment during the story, when everyone is rushing around trying to raise money for the foundation, John Jr. hands his father some money. He has sold his prized possession "Ripstik" to help his siblings.

The Zymagen Headquarters is located in Seattle, Washington. The Crowley family buy a house in Portland so they can be near to Zymagen and also to the hospital where Megan and Patrick get monitored and treated. Almost the entire movie was filmed in the Pacific Northwest around Portland. There are even some scenes that were filmed at the Nike Headquarters in Beaverton, Oregon.

Yep, SpongeBob Squarepants makes a cameo appearance in this great movie. Harrison Ford does an excellent job of appearing uncomfortable around the children during his visit to the Crowley residence. Earlier in the day Megan takes him outside for a running race up and down the sidewalk.

In a touching final scene, the parents and Dr. Stonehill are summoned to the hospital room by the sounds of laughter. Dr. Bob explains that the children are experiencing, for the first time, a "sugar high" which is making them giddy and happy. This behavior suggests that the enzyme is working.
